LAMA3 and amelogenesis imperfecta: On the other hand, in addition to AMTN and ODAM surfacing in the top 10 DEGs of P4-switch as compared to P4 organoids (Fig. 5c; Supplementary Dataset 6), laminin subunit gamma 2 (LAMC2) and laminin subunit alpha 3 (LAMA3), both expressed in mature ameloblasts in mouse incisor [46] and essential for amelogenesis in humans (with mutations linked to amelogenesis imperfecta [60, 61]), are highly upregulated in P4-switch organoids (Fig. 5c), further validated by RT-qPCR (Supplementary Fig. 5b).
