HLA-A and cancer: To identify and characterize these new antigens on cancer cells, originated by the arginine to histidine substitution on the residue 175 (R175H) in the TP53 gene [32], Hsiue and colleagues performed a positive selection phage display using naïve human antibody specific to HLA libraries, namely HLA-A*02:01 pHLA monomers, which loaded with the p53 R175H peptide, combined with negative selection against pHLA monomers loaded with the p53 wild type peptide.
