In human cancers, there are two common recurrent mutations in the TERT promoter region, which are located at two hotspots: chr5, 1,295,228 (GRCh37/hg19 by Entrez Gene) C>T (C228T) and 1,295,250 (GRCh37/hg19 by Entrez Gene) C>T (C250T), corresponding to the positions 124 and 146 bp upstream of the TERT translation start site, respectively [7].
