Though a series of Phase IIb screening trials suggested the PFS benefit for sorafenib plus capecitabine as first- or second-line treatment for patients with HER2-negative advanced breast cancer, the Phase III RESILIENCE trial showed the combination of sorafenib with capecitabine did not improve PFS, OS, or the overall response rate in patients with HER2-negative advanced breast cancer and the rates of Grade 3 toxicities were higher in the sorafenib arm (53, 54). The gene discussed is ERBB2; the disease is breast cancer.
