While APR-246 is being investigated in the Phase III clinical trial with TP53 mutant myelodysplastic syndromes (NCT03745716), one proposed mechanism of action is that both compounds are converted to methylene quinuclidinone (MQ) to covalently bind and modify thiols in the TP53 core domain, thus causes TP53 to restore its function to induce tumor cell death [32]. Here, TP53 is linked to myelodysplastic syndrome.
