In the course of tumor development, the volumes of the tumors were measured every 3 days from day 6 after injection and the result showed that tumor growth was remarkably decreased in both USP27 and SETD3 knockdown cells (Fig. 5A). At 27 days post-injection, tumors were dissected, photographed (Fig. 5B) and weighted (Fig. 5C). Tumors generated by USP27 or SETD3 knockdown cells were remarkably smaller and lighter than the tumors of control cells, while SETD3 overexpression could restore tumor growth in USP27-deficient cells.
