Zhang et al. [68] showed that TRAF6, a non-conventional E3 ligase, inhibited p53-mediated tumor suppression during genotoxic stress via an intriguing, two-step mechanism: (1) TRAF6 suppresses cytosolic p53 mitochondrial translocation via K63 ubiquitination thereby suppressing apoptosis and (2) TRAF6 ubiquitinates p53 at K24, thereby facilitating p53 engagement with p300, p53 acetylation, and anti-apoptotic upregulation of p21.
