2D monolayer U87 glioblastoma cells were cultured into 3D spheroid-forming conditions (neurospheres, Figure 2A), as these are known to be much closer to the tumor than adherent cells and to recapitulate some cancer stem cell phenotype as well as including increased expression of CD133, Nanog, and Sox2 (Figure 2B). The gene discussed is SOX2; the disease is cancer.
