For the third and final examined genome modification, we performed C-to-T conversion with cytosine base editors (CBEs)82 at the stop codon of the TYR loci, introducing a mutation known to cause albinism in the gene responsible for pig coat color.83,84 Sequencing-based analysis of 99 colonies indicated that 24.24% (24/99) of them were heterozygous, and 3.03% (3/99) were homozygous for the C-to-T base edit at TYR in the GN-pgEpiSCs background (termed GNT-pgEpiSCs) (Supplementary information, Fig. S10d, e). This evidence concerns the gene TYR and albinism.
