Figure 4E also includes a graph showing the effect of EWSR1-FLI1 knock-down on BrdU incorporation in the A673/TR/shEF cell line. EWSR1-FLI1 knock-down produced a 57% reduction in BrdU incorporation, greater than that observed after FEZF1 knock-down. Taken together, these results suggested that FEZF1 is involved in promoting cell proliferation in Ewing sarcoma cell lines. Interestingly, the effects of FEZF1 on cell proliferation were observed even in the presence of EWSR1-FLI1 expression (Figure 4A), suggesting that FEZF1 is a relevant downstream gene target in Ewing sarcoma oncogenesis. This evidence concerns the gene FEZF1 and Ewing sarcoma.
