TP53 and cancer: For each of these cancer genes, we identified the differentially essential genes between its WT and mutant cell lines in the CRISPR-Cas9 (AVANA) and shRNA (Achilles) screens, as described above for p53. We ranked the cancer genes by the significance of skewness in the numbers of differentially essential genes from CRISPR-Cas9 vs. shRNA screens similar to that shown in Fig. 1a for p53 (with Fisher’s exact tests, see the “Methods” section; results shown in Fig. 5a and Supplementary Data 6).
