Since the IL-6-Jak1-Stat3 pathway is a robust inducer of PC growth and metastasis, simultaneous suppression of Jak1-Stat3 is a desirable off-target effect of the Jak2 inhibitors in therapeutic applications for PC, which would expectedly lead to greater responsiveness of advanced PC to Jak2 inhibitor treatment. This evidence concerns the gene JAK2 and pachyonychia congenita.
