The function of PALB2 independent of BRCA1 in promoting DNA repair can impact the clinical outcome for cancer patients undergoing treatment with PARP1 inhibitors as it has been recently shown that restoring the function of PALB2 in BRCA1 null cancers also devoid of 53BP1 function can overcome resistance to PARP inhibitors (Belotserkovskaya et al., 2020). The gene discussed is PARP1; the disease is cancer.
