The orally available mammalian target of rapamycin (mTOR) inhibitors sirolimus and everolimus have variable absorption, often require therapeutic drug monitoring, and have incomplete target suppression.13-15nab-Sirolimus (nanoparticle albumin–bound sirolimus; ABI-009, formerly known as nab-rapamycin) is a novel intravenous (IV) mTOR inhibitor with significantly higher tumor growth inhibition, higher intratumoral drug accumulation, and greater mTOR target [phospho-S6 (pS6)] suppression compared with oral inhibitors, as demonstrated in preclinical models.16 This evidence concerns the gene MTOR and neoplasm.
