Phase III trial RTOG-1010 randomised 203 patients with HER2-positive adenocarcinoma involving the mid, distal oesophagus, or GOJ and up to 5 cm of the stomach, to chemotherapy (paclitaxel, carboplatin) and radiotherapy (50.4 Gy in 28 fractions) followed by surgery, or the same treatment with weekly trastuzumab prior to surgery and every 3 weeks for 39 weeks after the surgery. This evidence concerns the gene ERBB2 and adenocarcinoma.
