In a mechanistic study on how inhibition of the widely expressed transcriptional coactivator BRD4 leads to selective inhibition of the Myc oncogene in multiple myeloma, it was shown that BRD4 and mediator co-occupy in chromatin in a small set of exceptionally large super-enhancers associated with genes important for multiple myeloma. Here, BRD4 is linked to AL amyloidosis.
