We next examined the on-target inhibition of molecule 1, 14, and 20 in the human, pancreas-derived cancer lineage PANC1 as a proof-of-concept and used PANC1 cells, whether harboring an Akt1 knockout or an Akt2 knockout, as a cross validation for inhibitor induced decrease of downstream phosphorylation signals (Supplementary Fig. 15).
