Given the importance of STAG2 in cohesin function and its high rate of mutation in diverse types of cancer (Lawrence et al, 2014; Hill et al, 2016; Waldman, 2020), we performed genome-wide CRISPR-Cas9 knockout screens in three different genetic backgrounds to look for STAG2-dependent genetic interactions that could provide new insights into cohesin-STAG2 function and/or STAG2-based therapeutic strategies.
