The SNP rs249473 and especially the risk allele A, which lies within a silencer of the AKT1 locus (encoding for the AKT protein, part of the PI3K/AKT/mTOR signaling pathway), abrogates its silencing activity by creating a binding site for YY1, which in turn activates AKT1 transcription and elevates the risk of endometrial cancer [78].
