Approximately 80% of patients with Cowden syndrome have a germline inactivating mutation in PTEN. Kinston et al. reported an exceptional response to capivasertib in two cases with Cowden syndrome, breast cancer and germ-line PTEN mutations [260], indicating that Akt is a promising target for treating patients with germline PTEN mutations.
