As a use context and developmental proof of principle, we used the 96 × 96 barcoded primers and the three computational tools described here to identify and characterize mutants among thousands of Cas9-treated clones in the human adenocarcinoma cell line A549, specifically seeking disruptions of glucocorticoid receptor-occupied regions (GORs) near the glucocorticoid-responsive gene FKBP5 (Gencode v32 gene ENSG00000096060.14). The gene discussed is NR3C1; the disease is adenocarcinoma.
