T cells modified with transposons expressing tandem minigenes encoding multiple viral and tumor-associated epitopes connected by linker sequences elicited IFN-γ release from patient-derived CD8+ T cells reactive to an MHC class I–restricted viral epitope in CMV, CD8+ T cells isolated from a patient with lung adenocarcinoma reactive to a mutation in the PWP2 gene (45), and CD4+ T cells isolated from a patient with melanoma specific for the BRAF V600E mutation (ref. 46 and Figure 8C).
