ACSS2 and cancer: One of the reasons for interest in ACS is that there is no apparentphenotype in adult mice lacking ACSS2, indicating that this enzymeis not likely to be essential and, thus, raising its potential asa therapeutic target.4−7 Further supporting this concept is the fact that acss2–/– mice have a reduced tumor burden inmouse models of cancer.4,5 Similarly, Huang et al. have suggestedthat ACSS2 may also be a target for the treatment of fatty liver diseasebased on its role in the regulation of fat storage.8 Our interest in this pathway is also related to the nonessentialityof the pathway in mammals.
