Given the prevalence of MAPK pathway mutations in over 50% of newly-diagnosed myeloma patients [99], a trial investigating cobimetinib (cobimetinib, NCT03312530, Phase 1/2, multiple countries) enrolled 49 RRMM patients with median 4 prior lines of therapy in studies investigating the efficacy of MEK inhibition alone and combined with venetoclax (BCL-2 inhibition) and atezolizumab (PD-L1 inhibition) [32].
