While Tau is hyperphosphorylated in its disease-associated fibrillar state, phosphorylation sites (as well as other PTMs) were not observed in former cryo-EM structures of PHF-Tau from AD brains presumably due to PTM heterogeneity failing to provide a detail view of phosphate accommodation into filament assembly (Fitzpatrick et al., 2017).
