,7 Certain GPCs, such as GPC3 and GPC2, are oncofetal antigens that are expressed in early development and largely silenced in adult normal tissues but upregulated in epithelial solid tumors.7, 8, 9, 10 We and others have demonstrated that GPC2 is highly expressed in neuroblastoma (NB), one of the deadliest childhood cancers, and minimally expressed in normal tissues, making it an attractive candidate for CAR T cell therapy.11, 12, 13 Despite intensified cytotoxic therapy and immunotherapy, outcomes for patients with high-risk NB remain poor, with a long-term survival rate of <50%.14 This evidence concerns the gene GPC3 and neuroblastoma.
