AURKA and cancer: Starting from tozasertib (Figure 5), an Aurora A (AurA) inhibitor developed for cancer application, the use of computer-aided drug design to automate design and selection processes allowed the identification of pyrimidine 10 (Figure 5) with a 10,000-fold improved selectivity toward TrkA versus AurA, nanomolar cellular activity and high selectivity toward a large panel of protein kinases [20].
