A recent insight into this mechanism is provided by a study manifesting that in TRAIL-sensitive GBM cells, TRAIL-R2 is the only consistently expressed functional receptor that, upon TRAIL binding, homotrimerizes and recruits FADD and caspase-8 for assembling the DISC in the lipid rafts of plasma membrane, wherein caspase-8 is cleaved and initiates apoptosis. The gene discussed is TNFSF10; the disease is glioblastoma.
