In a pancreatic ductal adenocarcinoma (PDAC) mouse model, the inhibition of Sonic Hedgehog (Shh) in epithelial cells surprisingly results in tumor progression, in part reversed by a vascular endothelial growth factor receptor (VEGFR) blocking antibody, indicating that epithelial/stromal cells’ communication through Shh pathways promotes high stroma content in PDAC and acts to restrain the tumor at least in part through the inhibition of angiogenesis [123].
