Notably, the SIK inhibitor YKL-05-099 attenuated disease progression in two acute myeloid leukaemia (AML) mouse models, which was attributed to on-target inhibition of SIK3 based on overexpression of the SIK3 gatekeeper mutant in AML cells (Figure 1C) [127].
