To determine whether the changes imposed by acetylated tau on autophagy in the in vitro systems—namely, reduced CMA through disruption of lysosomal L2A dynamics and rerouting of acetylated tau oligomers to e-MI—were also observed in the brains of patients with Alzheimer’s disease (AD), we isolated two populations of lysosomes with different CMA activity, LE/MVB and autophagosomes from brains of 6 patients with autopsy-confirmed AD and 7 age-matched autopsy confirmed controls (no-AD) (patient information is summarized in “Methods” section).
