The basic structure of CAR-T cell has an extracellular domain consisting of a tumor-targeting/antigen-binding structure, i.e., an antibody derived single-chain variable fragment (scFVs); a hinge region; a transmembrane domain; and an intracellular signaling domain consisting of an activation motif with the CD3 zeta chain (T cell receptor complex), that provides signals for T cell activation leading to the production of toxic cytokines (e.g., IFNγ, TNFα) to attack tumor cells.
