AMG 510 significantly inhibits KRAS G12C and MAPK signaling in lung cancer cell lines but does not seem to influence wild-type KRAS. Initial results from a phase I/phase II clinical trial consisting of 533 patients with advanced NSCLC harboring KRAS G12C mutations as well as other solid tumors with KRAS G12C mutations has revealed that AMG 510 exerts a partial response in over 50% and a full response in 46% of the patients studied. The gene discussed is KRAS; the disease is non-small cell lung carcinoma.
