To date, studies investigating the roles of mammalian/mechanistic target of rapamycin (mTOR)16 and nuclear factor‐κB (NF‐κB)17 signalling pathways in immune dysregulation diseases have arguably a great effect in the inventories of mechanism‐based therapies, such as infliximab, an anti‐TNF agent in ameliorating autoinflammatory symptoms in patients with A20 haploinsufficiency,18 and sirolimus, an mTOR inhibitor used to treat activated phosphoinositide 3‐kinase δ syndrome (APDS).19 Here, MTOR is linked to Down syndrome.
