While CHD4 silencing or PAD1/3 expression did not alter overall PKM2 levels (Fig. 2f), strongly increased amounts of PKM2 were detected in the pan-citrulline IP following siCHD4 compared to siC in both 501Mel and MM117 melanoma cells and after ectopic PAD1 and PAD3 expression, particularly upon co-expression of both enzymes (Fig. 2g, h). This evidence concerns the gene PKM and melanoma.
