In a recent genome-wide association study (GWAS) meta-analysis of parental dementia and case-control Alzheimer’s dementia (ADem) [9], VKORC1 was associated (after Bonferroni correction) with ADem in a gene-based test (p = 5.1×10–8); the T-allele in rs9923231, which is related to the need for a lower dose of warfarin, was not a genome-wide significant finding, but was both located within a genome-wide significant locus and nominally associated with an increased risk of ADem (p = 1.8×10–7). Here, VKORC1 is linked to dementia.
