Knockdown of TETs by siRNA abolished the downregulation of fascin but not the upregulation of FILIP1L by vitamin C, while KDM5 inhibitor CPI-455 abolished the upregulation of FILIP1L, but not the downregulation of fascin by vitamin C (Figure 7D-I), indicating that KDM5 is likely responsible for the upregulation of FILIP1L and TETs largely mediate the downregulation of fascin by vitamin C. Overall, these results suggest that vitamin C improves the anti-tumor activity of buparlisib by cooperatively changing the expression of genes critical to TNBC growth and metastasis.
