Although new antiandrogens such as apalutamide and darolutamide continue to be developed and brought to the clinic, the emergence of tumour resistance due to point mutations and, more recently, the discovery of AR variants without the canonical druggable LBD which may contribute to resistance [79,80,108] (Figure 3) has contributed to the exploration of non-LBD centred pharmacological strategies.
