Definitive proof of the functional link between TNFR1 mediated necroptosis and neurodegeneration in MS-like pathology will require pharmacological intervention studies in our novel in vivo model, but when taken together with other evidence of necroptosis in MS [42], our data implies that targeting necroptosis could not only ameliorate TNF/TNFR1 induced neuronal cell death but also inhibit oligodendrocyte loss, thus having a dual action on demyelination and neurodegeneration. This evidence concerns the gene TNF and myeloid sarcoma.
