Here, in our more broad-based approach, we reasoned that molecular and cellular defects that are: (1) the most broadly common across the range of different mutations, and (2) the best correlated with the observed growth defects in the ded1-mam mutants, are the most likely to be critical functional changes both in yeast and, because of the functional conservation of DDX3X/Ded1, in medulloblastoma. The gene discussed is DDX3X; the disease is medulloblastoma.
