CLOVES syndrome belongs to a group of overgrowth syndromes that are collectively referred to as PIK3CA-related overgrowth spectrum (PROS) that are caused by somatic activating mutations in the gene encoding the phosphatidylinositol 3-kinase (PI3K) catalytic subunit alpha (PIK3CA) occurring early during embryogenesis3,4. The gene discussed is PIK3CA; the disease is CLOVES syndrome.
