The formation of GFP-LC3 puncta was observed in wild-type U2OS and human glioma H4 cells but not in cells that are deficient for the essential autophagy protein ATG5 and which acts upstream of LC3 to facilitate lipidation and membrane association (Fig. 1E, F and Supplementary Fig. 1).
