Temsirolimus, an mTOR inhibitor, and perifosine, an AKT inhibitor, were evaluated in a phase I study in 35 heavily pretreated patients with recurrent malignant gliomas (17 patients with rGBM) [173]; among the 29 evaluable patients, partial response was reported in one patient (3.4%) and stable disease in 13 patients (45%); mOS was 10.4 m (95% CI, 7.2–16.7) and mPFS was 2.7 m (95% CI, 1.8–9.2). This evidence concerns the gene AKT1 and malignant glioma.
