In AD, the Aβ-peptides that compose the senile plaques are derived from proteolysis of the amyloid peptide precursor protein (APP) in a coordinated sequence catalyzed by aspartyl protease beta-secretase 1 and 2 (BACE1/2) to generate the C-99 precursor that is processed by the presenilin-dependent γ-secretase [53].
