TP53 and cancer: While TP53 is one of the most frequently somatically mutated genes in human cancers (Olivier et al., 2010), with missense variants reported in more than 25 000 cancer samples in the Catalogue of Somatic Mutations in Cancer (Tate et al., 2019), somatic variants represent only 74 (11%) of the 649 missense VUS for TP53 in ClinVar.
