FAP and neoplasm: In distinct murine tumor models, multiple strategies targeting FAP exhibited therapeutic effects, including genetic deletion [24], pharmacological inhibition (PT630, PT-100) [25, 26], a novel monoclonal antibody (mAb) FAP5-DM1 [27], conditional ablation of FAP+ CAFs using diphtheria toxin [28] or αFAP-PE38 [29], and even novel FAP-targeting immunotherapies such as DNA vaccination [30], chimeric antigen receptor (CAR) T cells [31, 32] or oncolytic adenovirus [33, 34].
