CCL2 and neoplasm: The improved efficacy of the combination treatment when CCR2 antagonist and anti-PD-1 are used concurrently to target the tumor (Fig. 2) compared to CCL2 knockdown (Supplementary Fig. 4b) could be attributed to several reasons: better reduction in CCR2 receptor signaling that is achieved by reduction in CCL2; blockade of CCL2 being expressed by other cell types, such as CAFs, which are known to secrete CCL2 and promote macrophages in the tumor; and finally, non-specific effects of the inhibitor that serendipitously affect anti-PD-1 response.
