From genome-wide association studies carried out over a decade ago to more recent whole-genome sequencing projects of orofacial cleft cohorts, cleft-associated genetic loci continue to be identified, and the transcription factor IRF6 is one of the most commonly associated genes (Zucchero et al., 2004; Marazita, 2012; Yu et al., 2017; Cox et al., 2018).
