We found that two JAK/STAT pathway inhibitors, AG490 and Jaki, could reproducibly reactivate MeCP2. While the mechanism, efficacy, and safety of targeting this pathway must be thoroughly investigated, our results suggest that JAK-STAT inhibitors offer an additional approach that needs to be explored for the possible treatment of RTT and other X-linked disorders. The gene discussed is SOAT1; the disease is Rett syndrome.
