The impact of canonical Wnt/β-catenin signaling on bone formation and mechanoadaptation has been demonstrated, e.g., by the association of loss of function and gain of function mutations in the Wnt coreceptor low-density lipoprotein receptor related protein 5 (LRP5), which are associated with osteoporosis or high bone mass, respectively, in mice and humans [12,13,14].
